The enzymatic synthesis of psychosine.

چکیده
The ceramides active in this system have the three configuration and a triple or trans double bond. Ceramides containing the naturally occurring erythro isomers of sphingosine (2) and those with a cis double bond or no double bond are much less active. Since UDP-gala&se appeared to be the most probable precursor of the gala&se portion of cerebrosides, it has been postulated that cerebrosides might be synthesized by an analogous reaction C&3). Ceramide + UDP-galactose + cerebroside + UDP (2)

Versatile synthesis of phenoxydiazirine-based fatty acid analogues and photoreactive galactosylceramide.
A versatile synthesis of diazirine-based photoreactive fatty acid analogues is reported. The key step is phenoxy alkylation of diazirine with halo alkyl acid esters. The conditions described will be acceptable for the synthesis of various alkyl-length derivatives. The fatty acid derivatives are acceptors for reverse reactions of sphingolipid ceramide N-deacylase (SCDase), which catalyzes the co...

متن کاملKrabbe disease: psychosine-mediated activation of phospholipase A2 in oligodendrocyte cell death.
Globoid cell leukodystrophy (Krabbe disease) is an inherited neurological disorder caused by the pathogenomic accumulation of psychosine (galactosylsphingosine), a substrate for the deficient enzyme galactocerebroside beta-galactosidase. This study underscores the mechanism of action of psychosine in the regulation of oligodendrocyte cell death via the generation of lysophosphatidylcholine (LPC...
متن کاملGloboid cell leukodystrophy: deficiency of lactosyl ceramide beta-galactosidase.
Activity of lactosyl ceramide beta-galactosidase (beta-D-galactoside galactohydrolase, EC 3.2.1.23) was found to be extremely low in enzyme preparations from liver, brain, and cultured skin fibroblasts from patients with Krabbe's disease. Leukocytes from one set of parents had enzyme levels approximately half those measured in control leukocytes. The low activity observed for this galactolipid ...
متن کاملOptimization of Enzymatic Synthesis of Ampicillin Using Cross-Linked Aggregates of Penicillin G Acylase
Penicillin G acylase from E. coli TA1 was immobilized by Cross-Linked Enzyme Aggregates (CLEA), a new method for immobilization. This biocatalyst and commercial immobilized penicillin G acylase (PGA-450) were used to study the effect of pH, temperature and substrate concentration on the synthesis of ampicillin from phenyl glycine methyl ester (PGME) and 6-aminopenicillanic acid (6-APA). Compare...
